OPB-101 is an autologous, mesothelin (MSLN)-specific chimeric antigen receptor (CAR) T cell therapy developed for the treatment of solid tumors, particularly advanced platinum-resistant epithelial ovarian, fallopian tube, or primary peritoneal cancer. The therapy incorporates four proprietary technologies to enhance efficacy and safety: - An affinity and structurally optimized MSLN-targeted CAR for improved tumor targeting. - A novel regulated promoter (OUTLAST OP1) that mediates resistance to T cell exhaustion and localizes cytokine production at the tumor site. - A CD8α-targeted designed IL2/15 cytokine (OUTSMART IL2/15), which stimulates CD8-positive T cells and NK cells while avoiding regulatory T cell activation. - An EGFR-based safety switch (OUTSAFE EGFRopt), engineered for improved expression and sensitivity to clinically approved EGFR antibodies. These features are designed to promote robust CAR T cell expansion, persistence in the tumor microenvironment, enhanced anti-tumor activity, and increased safety by allowing controlled elimination of CAR T cells if needed[1][3][5][6][8].
