Overview

OPB-111077 is an orally bioavailable, small molecule inhibitor of signal transducer and activator of transcription 3 (STAT3) and mitochondrial oxidative phosphorylation. It was discovered and developed by Otsuka Pharmaceutical as a potential antineoplastic agent for the treatment of various cancers, including solid tumors and hematological malignancies such as acute myeloid leukemia (AML) and diffuse large B-cell lymphoma. The drug acts by inhibiting STAT3 phosphorylation, thereby blocking its activity both as a nuclear transcription factor involved in pro-tumorigenic gene regulation and as a regulator of mitochondrial oxidative phosphorylation. Preclinical studies demonstrated promising anticancer activity across multiple tumor types, but clinical trials have shown only modest efficacy in unselected advanced cancer populations[1][2][3][5][6][7].
