Overview

OPB-31121 is an orally bioavailable small molecule inhibitor targeting the signal transducer and activator of transcription 3 (STAT3) protein. STAT3 is a transcription factor involved in various oncogenic signaling pathways, making it an attractive target for cancer therapy. OPB-31121 binds with high affinity to the SH2 domain of STAT3, inhibiting its phosphorylation at key residues (pY705 and pS727), thereby blocking downstream signaling required for tumor cell proliferation and survival. The drug demonstrated promising preclinical antitumor activity and entered clinical trials for advanced solid tumors, liver cancer, leukemia, multiple myeloma, myelodysplastic syndromes, non-Hodgkin's lymphoma, and other cancers. Despite initial promise in early-phase studies, development was discontinued after phase I/II trials due to lack of sufficient efficacy or strategic reasons[1][2][3][4][5][6][7][8].
