Overview

OPC-28326 is a selective peripheral vasodilator developed as an antagonist of alpha2-adrenoceptors, with particular selectivity for the alpha2C-adrenoceptor subtype. It has been investigated primarily for use in peripheral vascular disease and Raynaud's phenomenon (including secondary Raynaud's in scleroderma). At low doses, it selectively increases femoral artery blood flow with minimal effects on systemic blood pressure or heart rate. The drug also demonstrates angiogenic activity in ischemic models and may mitigate postinfarction cardiac remodeling by increasing nonmyocyte proliferation and reducing apoptosis within infarct tissue. OPC-28326 does not significantly inhibit phosphodiesterase enzymes at relevant concentrations and shows only weak affinity for serotonin 5HT2 receptors at high concentrations[1][3][4][6][7].
