Overview

Ophiobolin A C21-methyl ketone is a synthetic derivative of the fungal metabolite ophiobolin A, developed to explore the structure-activity relationship of the C21-aldehyde group. Ophiobolin A, naturally produced by Bipolaris species, is known to induce paraptosis—a non-apoptotic form of programmed cell death—in glioblastoma (GBM) cells and glioma stem cells (GSCs). The C21-methyl ketone was synthesized by reacting ophiobolin A with Grignard reagents followed by oxidation, aiming to improve metabolic stability by replacing the potentially labile aldehyde group. However, research presented at SNO 2025 indicates that this modification results in inferior cytotoxic potency against patient-derived GSCs compared to the parent compound, suggesting that steric bulk at the C21 position is detrimental to activity.
