Overview

Opigolix (ASP1707) is an orally administered small molecule that acts as a gonadotropin-releasing hormone (GnRH) receptor antagonist. It was developed by Astellas Pharma for the treatment of endometriosis-associated pelvic pain and rheumatoid arthritis. By antagonizing the GnRH receptor, opigolix reduces levels of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), leading to decreased production of sex hormones such as estradiol. This mechanism aims to alleviate symptoms associated with estrogen-dependent conditions like endometriosis while minimizing hypoestrogenic side effects such as bone loss compared to traditional GnRH agonists[1][2][3][5][6][7].
