**OPL-0101** is a preclinical fusion protein therapeutic engineered to enhance CD8+ T cell function for cancer immunotherapy. Developed originally at Washington University in St. Louis and acquired by Valo Health via Courier Therapeutics in 2021, it binds with high affinity to NKG2D on cytotoxic lymphocytes while signaling through the common γ-chain cytokine receptor (IL-2R), separating binding and signaling to minimize off-target effects. This design improves T cell viability, memory formation, reduces exhaustion via NFAT pathway activation and JAK-3-mediated TCR phosphorylation, and outperforms IL-2/IL-15 in preclinical tumor models, synergizing with anti-CTLA4 for cures in lung cancer. It targets solid tumors by boosting low-affinity antigen recognition, with potential in melanoma, NSCLC, colon, and renal cancers; currently in IND-enabling studies.[1][3][5][6]
