OPN-9627 is an orally available, small molecule inhibitor of CD73 (ecto-5'-nucleotidase), developed by Opna Bio. It is designed to block the adenosine pathway in the tumor microenvironment, thereby preventing immune evasion by cancer cells. By inhibiting CD73, which catalyzes the conversion of AMP to immunosuppressive adenosine, OPN-9627 aims to enhance anti-tumor immune responses and may act synergistically with checkpoint inhibitors. The drug is being developed for gastrointestinal cancers including gastric, liver, pancreatic, and colorectal cancer (CRC). As of early 2025, it remains in preclinical or discovery stages[1][3].
