Overview

Opolimogene capmilisbac (JNJ-64041809, also known as ADU-741) is an investigational live attenuated double-deleted (LADD) *Listeria monocytogenes*-based therapeutic cancer vaccine. It was developed to target metastatic castration-resistant prostate cancer (mCRPC). The therapy encodes and expresses four antigens relevant to prostate cancer, aiming to stimulate the immune system against tumor cells. Upon intravenous administration, the engineered bacteria are taken up by antigen-presenting cells, leading to activation of both innate and adaptive immune responses. Clinical studies showed manageable safety but limited antigen-specific immune responses that did not translate into objective clinical benefit. Development was discontinued after early-phase trials.
