The **optimized arenavirus vector** is a genetically engineered, attenuated strain of lymphocytic choriomeningitis virus (LCMV), specifically WE-CL13 with mutations GP181M, GP185W, and GP492I in the glycoprotein, developed through serial passaging in tumor cells to enhance tumor tropism. It exhibits accelerated replication and propagation in various human tumor cells and organoids while showing limited replication in healthy cells, promoting strong, sustainable anti-tumor efficacy via T cell-mediated immunity without severe toxicity.
