Drug intelligence / Profile preview
optoRET is an optogenetic tool designed for the ligand-independent, light-controlled activation of the RET (REarranged during Transfection) receptor tyrosine kinase. Developed by researchers at the Korea Advanced Institute of Science and Technology (KAIST) and the Institute for Basic Science (IBS), optoRET is engineered by fusing the cytosolic domain of human RET with a blue-light-inducible homo-oligomerizing protein. Upon blue light illumination, optoRET undergoes clustering and autophosphorylation, recapitulating downstream RET signaling pathways including AKT and ERK. It has been utilized in research to study epithelial morphogenesis and branching in kidney organoids, as well as a preclinical gene therapy candidate to alleviate Parkinson's disease progression by promoting dopaminergic neuronal survival and behavioral recovery in mouse models.
