Overview

Opugotamig olatansine (IMGN-151) is a next-generation antibody-drug conjugate (ADC) targeting folate receptor alpha (FRα) designed to treat gynecologic and other solid tumors across a broad range of FRα expression levels.[1][3][8] It consists of a biparatopic anti-FRα monoclonal antibody conjugated via a protease-cleavable linker to the maytansinoid tubulin inhibitor payload DM21/DM21C, enabling efficient internalization and targeted cytotoxicity in FRα-expressing tumor cells while improving stability and exposure relative to first-generation FRα ADCs.[1][8][9][11] Developed initially by ImmunoGen and now in AbbVie’s pipeline, the drug is in early clinical development for recurrent ovarian, fallopian tube, primary peritoneal, endometrial, and cervical cancers, with intravenous administration in Phase 1/2 studies focused on safety, pharmacokinetics, and preliminary antitumor activity.[1][3][4][5][6][8]
