OR2H1-CAR T cells are chimeric antigen receptor (CAR) T cells engineered to target olfactory receptor family 2 subfamily H member 1 (OR2H1), an olfactory receptor overexpressed in various solid epithelial tumors, including non-small cell lung cancer (NSCLC), ovarian cancer, prostate cancer, and renal cancer, with expression in healthy tissues restricted primarily to the testes. Developed by researchers at the Moffitt Cancer Center, these CAR T cells utilize a single-chain variable fragment (scFv) targeting the extracellular domain of OR2H1 to selectively bind and eliminate OR2H1-expressing tumor cells. Preclinical studies have demonstrated significant in vitro and in vivo cytotoxic activity and tumor growth inhibition, particularly in chemotherapy-resistant ovarian cancer and NSCLC models, without causing toxicity to healthy tissues.
