OR502 is a fully humanized immunoglobulin G1 (IgG1) monoclonal antibody that targets the inhibitory immune checkpoint receptor leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2), also known as immunoglobulin-like transcript 4. By binding to LILRB2, OR502 reverses and prevents LILRB2-mediated immune suppression by myeloid cells and restores T cell functions, thereby enhancing both innate and adaptive anti-tumor immune responses. Preclinical studies demonstrated anti-tumor activity for OR502 alone and in combination with anti-PD-1 therapy. The drug is being developed primarily for advanced solid tumors, including cutaneous melanoma and non-small cell lung cancer (NSCLC). Clinical trials have shown an encouraging safety profile with promising early efficacy signals in monotherapy settings.
