Overview

OR641 is a novel, humanized bispecific monoclonal antibody that acts as a dual antagonist of leukocyte immunoglobulin-like receptor B1 (LILRB1) and leukocyte immunoglobulin-like receptor B2 (LILRB2). By binding to both LILRB1 and LILRB2 proteins, OR641 blocks their interactions with HLA class I ligands. This blockade relieves immunosuppression in the tumor microenvironment, promotes a Th1-like immune response, enhances IFN-γ secretion, decreases IL-10 secretion in stimulated peripheral blood mononuclear cells (PBMCs), modulates the function of M2-like tumor-associated macrophages (TAMs), and boosts adaptive anti-tumor responses. Preclinical data also show enhanced macrophage phagocytosis and increased NK cell-mediated cytotoxicity against lymphoma B-cells. The drug is being developed primarily for solid tumor malignancies as an immuno-oncology therapy[1][2].
