Drug intelligence / Profile preview
OraCOV is an orally administered, room-temperature stable vaccine candidate designed to protect against SARS-CoV-2 (COVID-19). Developed by Sanaria and Protein Potential in collaboration with the University of Tübingen, the vaccine utilizes the attenuated *Salmonella enterica* serovar Typhi strain Ty21a as a live bacterial vector. The Ty21a vector is genetically engineered to stably express and secrete SARS-CoV-2 antigens, specifically the spike ectodomain (eS) and the receptor-binding domain (RBD). To enhance survival in the acidic environment of the gastrointestinal tract, the vaccine incorporates a *Shigella sonnei* acid-resistance gene cassette. OraCOV is formulated as a foam-dried preparation that can be reconstituted in water and self-administered as a drink, inducing protective mucosal IgA, systemic IgG, and cellular T-cell immune responses.
