This herbal-based ORAI1 inhibitor is a natural product-derived therapeutic candidate being developed by Dongkwang for the treatment of atopic dermatitis. The drug's mechanism of action involves the inhibition of the ORAI1 ion channel, which is a key component of the store-operated calcium entry (SOCE) pathway. By blocking ORAI1, the drug aims to modulate calcium signaling in immune cells, thereby suppressing the production of pro-inflammatory cytokines and reducing the allergic inflammation characteristic of atopic dermatitis. The development of this compound was supported as a South Korean government research project from 2018 to 2022 and is currently in the preclinical stage.
