Overview

The oral polio vaccine (OPV) is a live attenuated viral vaccine administered orally to prevent poliomyelitis. Developed by Albert Sabin in 1961, OPV contains weakened forms of the three serotypes of poliovirus (types 1, 2, and 3), which induce immunity by replicating in the intestine and stimulating both mucosal and systemic immune responses. This mechanism not only protects vaccinated individuals but also helps interrupt person-to-person transmission of wild poliovirus within communities. OPV has played a critical role in reducing global cases of polio by over 99% since its introduction. However, rare cases of paralysis can occur due to reversion to neurovirulence or circulating vaccine-derived polioviruses (cVDPV). Novel versions such as nOPV2 have been developed for improved safety[1][3][5].
