OraPro-Zika is an orally administered, thermally stable vaccine candidate being developed by iosBio for the prevention of Zika virus infection. The vaccine is based on a non-replicating human adenovirus type 5 (AdHu5) vector (E1/E3 deleted) engineered to express the envelope (E) and non-structural protein 1 (NS1) of the Zika virus. It utilizes iosBio's proprietary OraPro platform technology, which incorporates a specialized formulation of sugars and modified amino acids to ensure thermal stability at temperatures up to 50°C. The vaccine is delivered via an enteric-coated capsule that protects the viral vector from gastric acidity, allowing it to reach the mucosal-associated lymphoid tissue (MALT) in the small intestine. This delivery mechanism is designed to trigger robust mucosal, systemic, and T-cell immune responses. Although development was approximately 90% complete and had shown promise in non-human primate trials, the program was paused in early 2020 to allow the company to focus on COVID-19 vaccine development.
