Overview

ORB-102 is a preclinical circular RNA (oRNA) therapeutic candidate developed by Orbital Therapeutics, which was acquired by Bristol Myers Squibb in December 2025. The drug is designed using Orbital's proprietary oRNA technology platform, which aims to provide durable and controllable protein expression by utilizing covalently closed RNA loops that are more stable than traditional linear mRNA. ORB-102 is specifically targeted for the treatment of Duchenne Muscular Dystrophy (DMD) by delivering a circular RNA payload that encodes a functional version of the dystrophin protein to muscle cells. This approach is intended to overcome the limitations of traditional gene therapies, such as the large size of the dystrophin gene and the challenges of redosing, by leveraging lipid nanoparticle (LNP) delivery for extra-hepatic targeting.
