ORB-PR1 is a lymphoid-targeted immunotherapy program developed by Orionis Biosciences, utilizing their proprietary A-Kine biologic and cell engager platforms. The program is designed to selectively activate CD8+ T cells and Natural Killer (NK) cells to elicit potent antitumor responses against solid tumors. ORB-PR1 includes a suite of molecules, such as CD8-targeted cytokines (specifically interleukins), engineered to provide localized immune activation within the tumor microenvironment while minimizing systemic toxicity. This approach aims to convert immunologically "cold" tumors into "hot" tumors, with preclinical models demonstrating significant efficacy and tolerability. As of early 2024, the program is in the IND-enabling phase of development, with multiple molecules progressing toward clinical trials.
