Orbitazine fumarate (ZX-101) is a novel small-molecule tubulin inhibitor developed by Shenzhen Zhenxing Pharmaceutical Technology Co., Ltd. for the treatment of high-grade gliomas, including glioblastoma multiforme. It exerts its therapeutic effect by binding to the colchicine-binding site on tubulin, which inhibits microtubule polymerization and disrupts the formation of the mitotic spindle. This disruption leads to cell cycle arrest at the G2/M phase and induces apoptosis in rapidly proliferating tumor cells. Orbitazine is specifically designed to possess high blood-brain barrier permeability, addressing a major challenge in the treatment of primary central nervous system malignancies. It is currently being evaluated in clinical trials for patients with recurrent or refractory glioma.
