SM-1 + temozolomide is an investigational combination therapy for the treatment of refractory or recurrent high-grade gliomas. SM-1 (orbitazine fumarate) is a novel small-molecule activator of procaspase-3 (PC-3), an enzyme that is frequently overexpressed in glioma cells. By directly activating procaspase-3, SM-1 functions as a proapoptotic agent to induce programmed cell death in tumor cells. Temozolomide is an oral alkylating agent that serves as the standard-of-care chemotherapy for glioblastoma by inducing DNA damage. The combination regimen aims to enhance the anti-tumor efficacy of chemotherapy through synergistic pro-apoptotic signaling. Developed by Shenzhen Zhenxing Medical Technology, the combination has been evaluated in Phase I and Phase II clinical trials in China.
