Overview

Orelabrutinib + obinutuzumab is a combination therapy under investigation for the treatment of B-cell malignancies, particularly marginal zone lymphoma (MZL) and chronic lymphocytic leukemia (CLL). Orelabrutinib is a highly selective, covalent Bruton’s tyrosine kinase (BTK) inhibitor that disrupts B-cell receptor signaling, leading to inhibition of malignant B-cell proliferation and survival. Obinutuzumab is a type II anti-CD20 monoclonal antibody that induces direct cell death and enhances antibody-dependent cellular cytotoxicity against CD20-expressing B cells. The combination aims to provide a chemotherapy-free regimen with high efficacy and manageable safety in previously untreated MZL patients. Preliminary clinical data show promising objective response rates and complete remission rates in first-line settings for MZL[2][7][9].
