Orelabrutinib + R-CHOP

Orelabrutinib + R-CHOP is a combination therapy consisting of orelabrutinib, a highly selective and irreversible Bruton tyrosine kinase (BTK) inhibitor, and the standard R-CHOP chemotherapy regimen (rituximab, cyclophosphamide, doxorubicin hydrochloride, vincristine, and prednisone). Orelabrutinib targets BTK to inhibit B-cell receptor signaling in malignant B cells with high selectivity and minimal off-target effects. The addition of orelabrutinib to R-CHOP has been investigated primarily for the treatment of diffuse large B-cell lymphoma (DLBCL), particularly in subtypes such as double-expression DLBCL or non-germinal center B cell–like DLBCL. Clinical studies have shown that this combination demonstrates promising efficacy and an acceptable safety profile in previously untreated patients with these forms of lymphoma[2][3][4][5][6][8].
