This combination therapy is a chemo-immunotherapy regimen consisting of orelabrutinib, zuberitamab, and bendamustine. Orelabrutinib is a highly selective, second-generation Bruton's tyrosine kinase (BTK) inhibitor that covalently binds to BTK, disrupting B-cell receptor signaling essential for the survival of malignant B-cells. Zuberitamab (HS-10370) is a recombinant humanized anti-CD20 monoclonal antibody that targets the CD20 antigen on B-cells, inducing cell death through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Bendamustine is a bifunctional nitrogen mustard alkylating agent that causes intra-strand and inter-strand DNA cross-links, leading to DNA damage and apoptosis. This triplet combination is primarily being investigated for the treatment of relapsed or refractory indolent B-cell non-Hodgkin lymphomas, including marginal zone lymphoma (MZL), follicular lymphoma (FL), and chronic lymphocytic leukemia (CLL), aiming to provide synergistic anti-tumor activity by simultaneously targeting multiple pathways in B-cell malignancies.
