Overview

Orexa (S1P-205) is a first-in-class, non-hormonal oral therapy being developed by S1 Biopharma for the treatment of male Hypoactive Sexual Desire Disorder (HSDD). It is a fixed-dose combination of two well-characterized antidepressants, bupropion and trazodone, formulated specifically to address the neurobiological imbalances associated with low sexual desire in men. Unlike treatments for erectile dysfunction that target peripheral vascular mechanisms, Orexa acts on the central nervous system to restore the natural balance of key neurotransmitters. Bupropion functions as a dopamine and norepinephrine reuptake inhibitor, enhancing excitatory signals in the brain's sexual centers, while trazodone acts primarily as a serotonin receptor antagonist (specifically targeting 5-HT2A and 5-HT2C) to reduce inhibitory signals. This dual approach aims to increase sexual desire by modulating the brain's reward and inhibitory pathways.
