Drug intelligence / Profile preview
ORF096 (also referred to as gpORF096) is a novel recombinant phage-derived structural protein originally identified as a component of the baseplate of therapeutic *Staphylococcus aureus* bacteriophages (such as A3R and 676Z). Developed by the Hirszfeld Institute of Immunology and Experimental Therapy, the Regional Specialist Hospital in Wrocław, and the Wrocław University of Science and Technology, ORF096 is being investigated as a precision antimicrobial agent against *Staphylococcus aureus*, including methicillin-resistant *S. aureus* (MRSA) strains. The protein structurally predisposes to form stable, biologically active trimers that exhibit potent lytic activity and cell wall degradation patterns. It targets the bacterial cell wall by binding to N-acetyl-D-glucosamine (NAG) and rhamnose substituents of wall teichoic acids (WTA), leading to rapid bactericidal effects and biofilm disruption without showing cytotoxicity toward mammalian cells.
