ORG-25435 is a synthetic small molecule drug developed by Organon International as an intravenous anesthetic agent. It acts as a positive allosteric modulator of the GABA A receptor and produces sedative effects comparable to propofol. The compound was designed as a water-soluble alternative to propofol and demonstrated rapid onset and recovery from anesthesia in clinical studies. However, its development was discontinued due to adverse effects such as hypotension and tachycardia at higher doses and unpredictable pharmacokinetics[1][2][3][5][6]. Its mechanism of action involves potentiation of GABA-induced chloride currents via allosteric modulation of the GABA A receptor[3][8].
