Overview

Org 25935 is a synthetic small molecule drug developed by Organon International (later acquired by Schering-Plough and then Merck) that acts as a selective inhibitor of the glycine transporter type 1 (GlyT-1). By inhibiting GlyT-1, Org 25935 increases extracellular glycine levels in the central nervous system. This mechanism was hypothesized to modulate glutamatergic neurotransmission and has been investigated for potential therapeutic effects in neuropsychiatric disorders. Preclinical studies showed that Org 25935 reduced alcohol consumption and exhibited analgesic and anticonvulsant properties. In clinical trials, it was primarily studied for its antipsychotic potential (notably as an adjunctive treatment for schizophrenia), prevention of relapse in alcohol dependence, and panic disorder. However, phase II trials did not demonstrate significant efficacy over placebo for these indications[1][2][3][5][6][7].
