Org 26576 is an experimental small molecule positive allosteric modulator of the alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptor. Originally developed by Organon and subsequently acquired by Merck Sharp & Dohme (via Schering-Plough), the compound was investigated for the treatment of major depressive disorder (MDD) and attention deficit hyperactivity disorder (ADHD). By modulating ionotropic AMPA-type glutamate receptors, Org 26576 enhances glutamatergic neurotransmission, a mechanism hypothesized to provide antidepressant effects by addressing glutamatergic dysfunction. Clinical development reached Phase 2, where it was evaluated for safety, tolerability, and maximum tolerated dose in patients with MDD. Despite demonstrating good tolerability and pharmacokinetic properties in early trials, clinical development of the compound appears to have been discontinued.
