Overview
ORG-34517 (also known as PT150 and SCH-900636) is a small molecule glucocorticoid receptor antagonist developed originally by Organon for the treatment of depression. It has also been investigated as an androgen receptor antagonist. The drug acts by blocking the glucocorticoid receptor, thereby normalizing hyperactivity in the hypothalamus-pituitary-adrenal (HPA) axis—a mechanism relevant to conditions characterized by excess corticosterone or cortisol activity. ORG-34517 has completed phase II clinical trials for major depressive disorder with psychotic features but development was discontinued for this indication. It is currently being developed by Pop Test Cortisol LLC/Pop Test Oncology for alcoholism and is in phase I clinical trials to evaluate its safety when taken concurrently with alcohol. Other potential indications explored include Cushing’s disease, hypertension, diabetes, and glaucoma[1][2][3][4][5].
