Overview

Ori-CAR-002 is a bispecific chimeric antigen receptor T cell (CAR-T) therapy developed by OriCell Therapeutics. It is engineered to simultaneously target two antigens—B-cell maturation antigen (BCMA) and G protein-coupled receptor class C group 5 member D (GPRC5D)—which are both highly expressed on multiple myeloma cells. The dual targeting approach aims to enhance anti-tumor efficacy and reduce the risk of tumor escape due to antigen loss. Preclinical studies demonstrated that Ori-CAR-002 exhibits robust proliferative responses, potent cytotoxicity against BCMA or GPRC5D-expressing cells, superior immune synapse formation, and complete tumor eradication in mouse models of multiple myeloma. The product was primarily developed for relapsed/refractory multiple myeloma, especially in patients who have failed prior BCMA-directed CAR-T therapies[1][4].
