Drug intelligence / Profile preview
Ori-TTV is an investigational in vivo CAR-T cell therapy candidate developed by Oricell Therapeutics. It utilizes a T cell-targeting lentiviral vector to directly engineer functional CAR T cells within the body, eliminating the need for ex vivo manipulation. Key innovations of Ori-TTV include an Ori Multiple Targeting Domain (Ori-MTD) for selective T cell transduction, a synthetic T cell-specific promoter for lineage-restricted CAR expression, a phagocytosis-resistant modification to evade macrophage clearance, and a de-targeted fusogen to enhance safety and reduce off-target effects. Ori-TTV encodes a dual-target CAR recognizing both CD19 and BCMA, facilitating the simultaneous elimination of malignant B cells and plasma cells. It is being developed for the treatment of hematologic malignancies (such as B-cell acute lymphoblastic leukemia, non-Hodgkin lymphoma, and multiple myeloma) and B/plasma cell-driven autoimmune disorders (such as systemic lupus erythematosus, idiopathic inflammatory myopathy, and systemic sclerosis).
