**ORIC-101 + nab-paclitaxel** is a combination regimen under clinical investigation for advanced solid tumors. ORIC-101 is a potent and selective small molecule antagonist of the glucocorticoid receptor (GR), aiming to reverse resistance to taxane-based chemotherapy. Nab-paclitaxel is an albumin-bound formulation of paclitaxel, a microtubule inhibitor widely used in cancer treatment. Preclinical data suggest GR signaling confers resistance to taxanes by upregulating antiapoptotic pathways; ORIC-101 blocks GR, potentially resensitizing tumors to taxanes. The combination has been studied in phase 1/1b trials primarily in patients with advanced or metastatic solid tumors refractory to taxane-based regimens, including pancreatic, ovarian, triple-negative breast, and other tissue-agnostic cancers. Although ORIC-101 achieved clear GR pathway inhibition and the regimen was tolerable, clinical benefit was limited in taxane-resistant populations, with objective response rates insufficient to justify continued development in some trials[1][3][5].
