Overview

ORIC-533 is a highly potent, orally bioavailable small molecule inhibitor of CD73, an enzyme that plays a central role in the adenosine pathway. By binding to and inhibiting CD73, ORIC-533 prevents the conversion of adenosine monophosphate (AMP) to adenosine, thereby reducing immunosuppressive adenosine levels in the tumor microenvironment. This mechanism restores and enhances immune function by counteracting tumor-driven immunosuppression. ORIC-533 is being developed by ORIC Pharmaceuticals for the treatment of relapsed/refractory multiple myeloma and has demonstrated strong target engagement, favorable pharmacokinetics (plasma half-life ~24 hours), and preliminary clinical activity in early-phase trials[1][2][3][4][5][6].
