OriC321 is an investigational chimeric antigen receptor T cell (CAR-T) therapy developed by OriCell Therapeutics in collaboration with Zhejiang University. It is designed as a dual-targeted CAR-T product that simultaneously targets B-cell maturation antigen (BCMA) and G protein-coupled receptor class C group 5 member D (GPRC5D), both of which are highly expressed on malignant plasma cells in multiple myeloma. The therapy uses a proprietary signal activation domain to enhance the expansion, persistence, and anti-tumor activity of the engineered T cells within the tumor microenvironment. Its primary indication is for relapsed or refractory multiple myeloma, aiming to overcome resistance mechanisms seen with single-antigen targeting therapies[1][2][3][5].
