OriC613 is a dual-target CAR-T cell therapy developed by Oricell Therapeutics Co., Ltd. It is designed to treat solid tumors, specifically pancreatic and gastric cancers, by targeting both Mesothelin (MSLN) and Claudin 18.2 (CLDN18.2). The therapy utilizes an "AND" logic gate design, incorporating an anti-CLDN18.2 scFv with high binding strength and an anti-MSLN VHH with moderate binding affinity. This design ensures full activation exclusively in tumor cells that co-express both MSLN and CLDN18.2, aiming to enhance anti-tumor efficacy while mitigating off-target gastrointestinal toxicity associated with targeting CLDN18.2 alone.
