OrienX010 is an oncolytic virus therapy derived from herpes simplex virus type 1 (HSV-1), genetically engineered to express human granulocyte-macrophage colony-stimulating factor (GM-CSF). The virus has been modified by deleting or inactivating three viral genes (ICP34.5, ICP47, and ICP6) to enhance tumor selectivity and safety. Deletion of ICP34.5 allows selective replication in cancer cells while reducing neurovirulence; deletion of ICP47 enhances antigen presentation and T cell priming; inactivation of ICP6 further reduces neurotoxicity risk. The insertion of the GM-CSF gene aims to stimulate local immune responses against tumors. OrienX010 is administered via intratumoral injection and is being developed primarily for the treatment of unresectable or metastatic melanoma[1][2][4].
