ORL-101 is an investigational **pharmaceutical-grade ultra-pure l-fucose** small-molecule therapy being developed by **Orpha Labs** for **Leukocyte Adhesion Deficiency Type II**. LAD-II is caused by defective fucosylation, and ORL-101 is intended to restore or improve the fucosylation of cell-surface and plasma membrane glycoproteins, including **E-selectin and P-selectin ligands**, thereby addressing the core biochemical defect underlying impaired leukocyte adhesion. The program has been described as cGMP-manufactured and has received FDA **Orphan Drug**, **Rare Pediatric Disease**, and **Fast Track** designations; a Phase III trial was initiated in 2022, and the drug was also made available through compassionate use starting in 2021.
