Overview

Orludodstat (BAY 2402234) is a novel, potent, and selective small molecule inhibitor of dihydroorotate dehydrogenase (DHODH), an enzyme involved in the de novo pyrimidine synthesis pathway. By inhibiting DHODH, orludodstat induces differentiation and apoptosis in acute myeloid leukemia (AML) cells and demonstrates efficacy across multiple AML subtypes. It has shown strong anti-proliferative activity in vitro against AML cell lines at low nanomolar concentrations and induces cell cycle arrest at the G2/M phase. In vivo studies have demonstrated reduced tumor volume and increased survival in mouse models of AML. Orludodstat is being developed primarily for the treatment of myeloid malignancies such as AML, myelodysplastic syndrome (MDS), chronic myelomonocytic leukemia (CMML), and has also been investigated for recurrent glioma[1][2][3][4][5][6][7].
