ORM-12741 is an orally available small molecule that acts as a potent, selective antagonist of the alpha-2C adrenergic receptor. It was developed by Orion Pharma primarily for the treatment of Alzheimer's disease. The drug targets a specific subtype of adrenergic receptors (alpha-2C) in the brain, which are believed to modulate brain functions under stressful conditions and may play a role in cognitive deficits associated with Alzheimer's disease. ORM-12741 has demonstrated high affinity and selectivity for the human alpha-2C adrenergic receptor over other subtypes (alpha-2A and alpha-2B), with functional antagonism confirmed in vitro[1][6]. In clinical trials, it has shown statistically significant benefits on memory performance and neuropsychiatric symptoms when used as an add-on therapy to standard cholinesterase inhibitors or memantine in patients with moderate Alzheimer's disease[1][5]. The compound was originally part of a schizophrenia drug discovery program but is no longer being developed for that indication[4]. Its pharmacokinetic properties make it suitable for oral administration.
