**Orosartan** is an oral fixed-dose antihypertensive combination of amlodipine orotate and valsartan, originally developed under the code **G-0081** by Dong-A Pharmaceutical and subsequently developed and marketed by Dong-A ST in South Korea. Amlodipine inhibits vascular L-type calcium channels, producing arterial vasodilation, while valsartan selectively antagonizes the angiotensin II type 1 receptor to reduce angiotensin II-mediated vasoconstriction and aldosterone signaling. G-0081 was evaluated against Exforge in a Phase 1 pharmacokinetic study and Orosartan was subsequently studied in essential hypertension.
