ORP-100 (also known as OrPro and later rebranded as Theradux) is an experimental biologic drug developed to improve mucociliary clearance in the airways, particularly for patients with cystic fibrosis. The drug is designed to enhance mucus transport by modulating the properties of airway mucus, thereby facilitating its removal from the respiratory tract. It is a modified form of thioredoxin engineered for increased activity and stability when delivered directly to airway surfaces via inhalation. This modification addresses limitations of native thioredoxin such as poor half-life and undesirable cellular uptake. Preclinical studies have shown that ORP-100 can reduce abnormal stiffness and viscosity in cystic fibrosis sputum, restore normal mucociliary transport rates on tracheal surfaces, and modulate pro-inflammatory cytokine release both in vitro and in vivo[1][9].
