Overview

ORP-101 is a new chemical entity and small molecule drug developed by OrphoMed for the treatment of irritable bowel syndrome with diarrhea (IBS-D). It acts as a peripheral partial agonist at the μ-opioid receptor and a full antagonist at the κ-opioid receptor. This dual mechanism is designed to address both dysmotility and pain symptoms in IBS-D without causing central nervous system effects or risks such as sphincter of Oddi spasm or pancreatitis. ORP-101 does not cross the blood-brain barrier, making it likely non-addictive. The drug has completed Phase 2 clinical trials and was granted FDA Fast Track designation in 2018[1][4][5][6].
