ORTD-1

ORTD-1 (also known as Orynotide ORTD-1 or RTD-1) is a synthetic macrocyclic peptide based on rhesus theta-defensin 1. It is being developed by Oryn Therapeutics as a novel immunomodulatory therapy for systemic inflammatory diseases including rheumatoid arthritis and COVID‑19-related pneumonia. Mechanistically, it acts as an immunomodulator with inhibitory effects on ADAM17 (a disintegrin and metalloproteinase domain-containing protein 17) and the NF-kappa B pathway. Preclinical data suggest it is non-toxic and non-immunogenic with favorable pharmacokinetics[2][4][9]. Clinical development has included phase 1 studies in rheumatoid arthritis and phase 2 studies in COVID‑19 pneumonia; however, further development appears to have been discontinued[2][5].
