**orthoIL-2 1G12** is an engineered variant of the cytokine interleukin-2 (IL-2), designed to interact selectively with a mutated “orthogonal” IL-2 receptor beta (orthoIL-2Rβ) subunit expressed on specifically engineered T cells. Unlike wild-type IL-2, orthoIL-2 1G12 does not activate natural IL-2 receptor-expressing cells, resulting in highly restricted immune modulation. It maintains potent activation of orthoIL-2Rβ-transduced T cells (effector and regulatory T cell subsets), driving expansion and function of adoptively transferred, engineered T cells with reduced off-target toxicity and minimal activity on host wild-type cells at specific doses. This makes orthoIL-2 1G12 a synthetic immunotherapy tool for cell therapy—especially in adoptive cell transfer protocols and cancer models—improving anti-tumor responses while minimizing systemic immune activation and toxicity. Its specificity is based on engineered protein-protein interactions and does not broadly stimulate endogenous immune populations[1][3][5].
