Orthokine (also known as Regenokine in the United States) is an autologous conditioned serum (ACS) developed by Orthogen AG. It is prepared by incubating the patient's own whole blood with medical-grade chromium sulfate-treated glass beads, which mechanically stimulates white blood cells to produce high concentrations of anti-inflammatory cytokines and growth factors, particularly interleukin-1 receptor antagonist (IL-1Ra), along with IL-4, IL-10, and IL-13. The resulting cell-free serum is separated via centrifugation and administered via intra-articular or perineural injection. Orthokine is primarily indicated for the treatment of joint and spinal disorders, including knee osteoarthritis and degenerative back pain, where it acts by blocking the pro-inflammatory IL-1 pathway to reduce pain, alleviate joint inflammation, and slow cartilage degradation.
