orthoPmel is an experimental adoptive T-cell therapy (ACT) consisting of T cells derived from pmel-1 T cell receptor-transgenic mice that have been engineered to express a mouse orthogonal IL-2 receptor beta (moRβ). Developed by Synthekine, this system is designed to selectively respond to a specific pegylated IL-2 mutein (moIL-2) that does not activate wild-type IL-2 receptors. This orthogonal signaling platform allows for the selective expansion and activation of the transferred T cells within the tumor microenvironment without the systemic toxicities associated with high-dose IL-2 or the requirement for pre-conditioning lymphodepletion. In preclinical B16 melanoma models, orthoPmel cells demonstrated significant systemic and intratumoral expansion and controlled tumor growth when combined with moIL-2.
