ORY-3001 is a highly potent and selective oral small molecule inhibitor of lysine-specific demethylase 1 (LSD1), an epigenetic enzyme that removes methyl groups from lysine 4 of histone H3, thereby regulating gene transcription. Developed by Oryzon Genomics, ORY-3001 is designed for non-oncological indications, with preclinical studies demonstrating its ability to increase fetal hemoglobin (HbF) levels by upregulating γ-globin gene expression in animal models of sickle cell disease (SCD). The drug has shown efficacy in increasing F reticulocytes, γ-globin chain synthesis, and γ-globin mRNA in both SCD mice and baboons. Its mechanism targets LSD1 to modulate chromatin structure and promote beneficial changes in gene expression relevant to hemoglobinopathies[1][2][3][4][5].
